How to Filter Buffers (Vacuum Filtration)

In this video, Dan shows you how to properly assemble and use the vacuum filtration apparatus. Sometimes you need to filter your buffers or other solutions before you use them. If you are using any automated liquid chromatography systems (such as AKTA or FPLC systems), then you need to filter your buffers before loading them on the columns. Remember: TO START: Attach tubing THEN turn on tap. TO END: Remove tubing THEN turn off tap. *If you have detergent in your buffer, bubbles will form during filtration -- make sure to continuously monitor and break the vacuum before the bubbles rise up to the level of the tubing.
